Pyridoxal-5-phosphate Monohydrate is the cofactor of this enzyme and the normal coenzyme form of Vitamin B6.

Pyridoxal phosphate (PLP, pyridoxal-5'-phosphate, P5P), the active form of vitamin B6, is a coenzyme in a variety of enzyme reactions. The Enzyme Commission has catalogued more than 140 activities that rely on pyridoxal-5-Phosphate, equivalent to ≈4% of all classified activities. The versatility of pyridoxal-5-Phosphate stems from its ability to covalently bind substrates and then act as an electrophilic catalyst, thereby stabilizing different types of carbon anion reaction intermediates.

Pyridoxal-5-Phosphate is the active form of vitamin B6 in the body, that is, a coenzyme for transaminases, decarboxylases, cysteine desulfurase, etc., and plays a very important role in the metabolism of proteins or amino acids. Pyridoxal-5-Phosphate is a coenzyme for transaminases and decarboxylases in amino acid metabolism. It is currently widely used in microbial fermentation as a nutrient for microorganisms. The actions of Pyridoxal-5-Phosphate are mainly related to the metabolism of amino acids and other nitrogen compounds:

1: In the liver, Pyridoxal-5-phosphate is required for glucose production from amino acids through its role as a coenzyme of transaminases. Pyridoxal-5-phosphate is also required in the liver and muscles to obtain stored glycogen for glucose production and to synthesize niacin from the amino acid tryptophan.

2: In erythrocytes, Pyridoxal-5-phosphate is required for hemoglobin synthesis and regulation of oxygen affinity. In addition, cellular receptivity to steroid hormones is affected by Pyridoxal-5-phosphate.

3: In the nervous system, Pyridoxal-5-phosphate is required for the synthesis of several neurotransmitters, including 5-hydroxytryptamine (from tryptophan), taurine, dopamine, norepinephrine, histamine, and gamma-aminobutyric acid (GABA). Many experimental and clinical observations support the fact that dietary vitamin B6 is required to maintain normal nervous system function.

4: Together with vitamin B12 and folic acid, Pyridoxal-5-phosphate is involved in single carbon metabolism and is necessary for maintaining normal plasma homocysteine levels. Elevated plasma homocysteine may be an independent risk factor for the development of cardiovascular disease. In addition, due to its participation in single carbon metabolism, Pyridoxal-5-phosphate helps maintain normal immune function.
