delta-Valerolactone is a versatile fine chemical intermediate widely used in pharmaceutical intermediates (such as in the synthesis of pyrimidinopyrimidines, cilostazol, etc.), fragrances and flavors, polyesters, polyurethanes, specialty solvents, and coatings.
1: delta-Valerolactone and cyclohexylamine are used as raw materials to synthesize N-cyclohexyl-5-chlorobutyl-1H-tetrazole, an important intermediate for the antiplatelet drug cilostazol. delta-Cyclopentanolide is also an intermediate in the synthesis of the antifertility drug (+)-Phoiboxazole. The 2,3-dihydrofuran produced by the reduction reaction of delta-cyclopentanolide can be used to synthesize prostaglandin analogs such as misoprostol and the uterotonic agent dinoprostone.
2: delta-Valerolactone can be used to synthesize optically active (+)-Quebrachamine cyclic ethers, which are a large class of bioactive natural tricyclic compounds.
3: delta-Valerolactone reacts with nitriles in the presence of trifluoroacetic anhydride to form 2,4-disubstituted pyrano[2,3-d]pyrimidines, exhibiting antibacterial activity and potential use as bactericides and herbicides.
4: delta-Valerolactone undergoes an ester condensation reaction with ethyl trifluoroacetate to produce trifluoroacetyl-delta-valerolactone. This compound plays a significant interdisciplinary role in fluorochemical research and has broad application value.
5: delta-Valerolactone readily undergoes ring-opening polymerization, with extensive research focused on the modification of polymeric materials, particularly in the fields of medicine and liquid crystals. delta-Valerolactone itself easily polymerizes to synthesize poly(δ-valerolactone). This polymer exhibits biocompatibility and biodegradability, making it highly relevant in areas such as biomedical materials, biodegradable plastics, and high-value-added packaging materials. delta-Valerolactone can also be copolymerized with other monomers to form copolymers with diverse properties. Ring-opening polymerization of delta-valerolactone with other lactones (such as caprolactone) produces polylactones that are biodegradable, soft, and transparent, making them suitable for treating various wounds in medical applications due to their good absorbability and strong adhesion.
6: delta-Valerolactone can also be used to synthesize environmentally friendly plastics and absorbable sutures for surgical procedures. Polymers of cyclopentanolide can also be used topically on human skin in products such as sunscreens and cosmetics.
7: delta-Valerolactone can be used to synthesize formulations for modifying wood materials, to prepare a resin for coatings on electrical appliances, and as a non-aqueous electrolyte for capacitors in electrochemistry. The ammonolysis of delta-valerolactone with aromatic diamines yields aromatic ammonium salts, which upon polycondensation form hydrogen-bonded aromatic and aliphatic polyamide esters. These polymers are molding materials for pressure glazing and exhibit thermal stability. It can also be used to synthesize optically active substances that serve as raw materials for liquid crystal materials. Utilizing the high surface tension of cyclopentanolide, it can be employed as a solvent for the synthesis of oligoribonucleotides and oligopeptides.
